Physical obstructions such as planets, stars, gas, and dust would be a factor as well as gravitational fields from those objects. Then, you'd also have to have a near-perfect aiming & timing system to hit a target a few light-years away. It would be a pain to have to aim at a point in space where the target will be a few years later when the laser finally gets there.

I highly, highly doubt that life on Earth is the only life humans will encounter during their existence, but I wouldn't be surprised in the slightest if we never encountered intelligent life as we know it - technology-bearing, abstract-thinking social animals.
