The reclusive Borat star says he's become too well-known to get away with it.
And he admits that pretending to be the offensive Kazakhstani reporter for weeks on end left him exhausted.
In a rare interview out of character, the English-born comedian told US radio that his next project would have to be something completely different.
He said: "I think it will probably be impossible.
"There are a number of things that are in development.
"The [next thing] will probably be a scripted comedy.
"I think the days of me going undercover are probably over."
He added: "There are a number of challenges.
"One is that I have to stay in character usually for about eight hours at a time, because if someone gets suspicious and says, 'Wait a minute, he's standing there in the corner like a guy from England', then everything's over.
"There's a lot of physical preparation - I never washed the suit, when I come into the room there's immediately this terrible smell. Borat doesn't wear deodorant so there is this kind of dreadful Soviet-bloc smell the moment I walk in."
But movie insiders said last night his comments could put a multi-million dollar deal with one of Hollywood's biggest studios at jeopardy.
The actor, 35, reportedly signed a US$42.5m film deal starring his comic creation Bruno.
According to Hollywood Reporter, a deal has been made with Universal Pictures to star as the gay Austrian fashion obsessive. It is thought the movie would be similar to Borat, with Cohen going undercover to infiltrate the world of haute couture.
A Hollywood insider said: "They were banking on him coming up with another Borat.
"There's no guarantee people will flock to see something that is scripted.
"Whatever he does next could make or break his career in Hollywood."
And he revealed he sometimes felt like an assassin when he was making the film - especially during a dinner party scene where he returns to the table from the toilet clutching a bag of his own human waste.
He added: "There is a moment when you feel almost like a hit man.
"You look in the mirror and it's as if you've pulled the gun out and you're about to assassinate someone and you're doing a hit and you know everything's going to change. Everyone's having a good time but I'm about to bring a bag of fresh faeces to the table."
He told US radio station NPR [National Public Radio]: "At some point I've got to reveal the real me, reveal the real moustache."
And he laughed off speculation that he could be in line for an Oscar at this year's Academy Awards.
He added: "It would be so bizarre.
"When I made this film I never intended to get any critical acclaim.
"You want to make your friends and fans laugh. I'm just happy to get a couple of nominations and return to England."
It comes as the spoof documentary, Borat: Cultural Learnings Of America For Make Benefit Glorious Nation Of Kazakhstan was nominated for a prestigious Writer's Guild of America (WGA) Award for Best Adapted Screenplay at the weekend.
Cohen is also in the running for a Golden Globe as Best Actor in a Musical or Comedy.

Although, in reality It would be great if he did win, just to show good acting isn't always about doing serious (usually seriously boring) dramas. When Baron pretty much moved to the US after becoming too well known across the rest of the world, I always wondered how he would be able to continue playing his characters when he became well-known there too (seeing as he plays off unsuspecting victims). I guess instead of going to new markets now, he's just going to continue inventing new characters (which is better IMHO).
